1/10 – Monday –  B Day

Main focus –   
                                                                                                                                                                                     a)  To complete the graphs of nucleotide base pair lengths from the Crime Scene                         
b)  To use an exponential  graph
c)  To begin the PCT paper PCR lab – isolate the DNA


1: Continue the measuring of the nucleotide base pairs from our Crime Scene gels.

2: Begin making a graph of base pair length and distance measured. – make a best fit line.

3. Use the best fit line to obtain the base pair values for you bands in the gel.

PTC Taster PCR Lab Data:
How to understand the Gels: 


The idea is very simple. If you are a taster that means you have at least one chromosome that has the palindrome that the restriction enzyme can cut intact and thus you should have smallest fragments (44 base pairs)  visible and the 177 fragment visible (middle band).  

The largest fragment (221) will be visible if at least one chromosome HAS the mutation in the palindrome region and thus the restriction enzyme could not cut the DNA. This band will identify a non-taster.

You will start your Genetics Test Tomorrow with Multiple Choice and         vocabulary.  There is Genetics Review Page That I have posted in 2nd quarter.  There is a lot there             BUT remember you have been studying everyday by completing every assignment.  This test is                  really about the big overlying concepts.       
  – Please review your Practice AP questions that I have given you as well.
