3. Complete the Drosophila Lab with packet. – We did not get to do this today

1/12 – Tuesday – “B” Day – period 7B, 8B– I 7(B) 8(B,D) AP BIOLOGY – (double period Lab)

Period 8,9
1: pGlo Transformation Lab-
a) obtain 2 micro test tubes, one labeled +pGlo and the other -pGlo
Little p means plasmid – small circular DNA of E. Coli.
b) Obtain your starter plates that you streaked yesterday that has been incubating for 24 hours at 37 degrees
Celsius.
c) Use a sterile loop and pick up a single colony and disperse into each tube, +pGLO and -pGlo.
d) I will add 10 micro liters of the pGlo plasmid (engineered sequence) to your +pGLO micro test tube.
e) Incubate the tubes on ice for 10 minutes.
f) label your lids.
g) heat shock, add broth, pippete and streak in the appropriate plates.
f) Stack up the plates, tape and label, then incubate for 24 hours.

1. Complete the Genetics Test Homework list with definitions from your text using the vocabulary words posted above. You will take the Vocabulary part of the test next Wednesday/Thursday.
2. Complete the pGlo Lab packet up to page 16. I will collect Tuesday and Wednesday next week.
3. Complete the following pages in the Crime Scene Lab:

You need a gel that has excellent clarity to answer some of these questions. Use any gel from
either class if you own gel that I posted here does not have good clarity.
